چکیده

Intact human red blood cells can synthesize FAD and FMN from riboflavin. The rate of synthesis of FAD is linearly proportional to the concentration of riboflavin in the medium at levels below 0.9 1 M. With 0.9 1 M riboflavin, the rate of synthesis is about 0.1 m mole FAD/ml. red blood cells/hour. Incubation of red blood cells with riboflavin can result in increased red cell glutathione reductase activity when the enzyme is measured in the absence of added FAD. This mdicates that the FAD concentration in the red cells increased during the incubation. The rate of incorporation of radioactive riboflavin into red blood cells is the same whether the cells are suspended in plasma or in a phosphate-saline-glucose medium. The time it takes for half the FAD in normal human red blood cells to turn over is calculated to be about 6 days, assuming a single mixing pool of red cell FAD.

Application of Riboflavin-Deficient Rat Erythrocytes in the in Vitro Assay of Faba Bean Toxicity

Background: Faba bean toxicity (favism) is a common condition observed in our region. Objective: To develop a short and simple technique involving the use of riboflavin-deficient rats to test in vivo faba bean toxicity. Methods/Results: Sprague Dawley rats were maintained on a riboflavin-deficient diet and their vitamin B2 status was monitored by the assay of erythrocyte glutathione reductase (...

Enzymatic deficiencies of purine nucleotide synthesis in the human erythrocyte.

In the development of the mature rabbit erythrocyte from the reticulocyte, the capacity for the synthesis de novo of the purine ring in vitro is lost (2, 3). The mature rabbit erythrocyte, however, does retain a portion of the biosynthetic pathway de novo, as evidenced by its utilization of 5-amino-1-ribosylWimidazolecarboxamide and sodium formate-Cl4 for the formation of inosinic acid (4). The...

Synthesis of Two Compounds with Self-Assembled Monolayer Properties: Riboflavin 2', 3', 4' , 5' Tetra Octadecanoate & Bis (Phosphatidyl Ethanol) Protoporphyrin IX Amide

Riboflavin and protoporphyrin IX are two molecules that participate in oxidation and reduction reactions in the living cell. Changing some functional groups of riboflavin and protoporphyrin IX can provide compounds with self-assembled monolayer properties with wide applications in designing the molecular electronic devices. In this study, the amphiphilic structure of riboflavin and protopor...

Glutathione peroxidase (EC 1.11.1.9) and superoxide dismutase (EC 1.15.1.1) activities in riboflavin-deficient rats infected with Plasmodium berghei malaria.

Riboflavin deficiency interferes with the growth and multiplication of malaria parasites as well as the host response to malaria. The objective of the present work was to determine the effects of riboflavin deficiency on erythrocyte glutathione peroxidase (EC 1.11.1.9; GPx) and superoxide dismutase (EC 1.15.1.1; SOD) in rats infected with Plasmodium berghei malaria. Riboflavin in its co-enzyme ...

Enzymatic synthesis of the pteridine moiety of dihydrofolate from guanine nucleotides.

Much of the knowledge of pteridine biosynthesis from purines has come from radioactive tracer studies with whole organisms. Reports on the incorporation of the radioactivity of labeled precursors of purines (1) and of uniformly labeled adenine-1% (2) into riboflavin indicated the utilization of purines for riboflavin synthesis. Furthermore, the results of studies (2, 3) with adenine-814C showed...
